Malarone Paediatric 62.5 mg/25 mg film-coated tablets are a prescription-only antimalarial medicine used for the prevention of malaria in children.
Each tablet contains atovaquone and proguanil hydrochloride, a combination that provides protection against malaria caused by Plasmodium falciparum, including strains resistant to chloroquine.
This paediatric formulation is licensed for use in children weighing 11 kg to 40 kg and is taken once daily with food.
Dosing in children weighing 5 kg to 11 kg is outside the licensed indication and follows current UKHSA guidance from the Advisory Committee on Malaria Prevention.
Malarone Paediatric is suitable for short trips and longer stays in malaria-endemic areas and requires a short post-travel dosing period of seven days after leaving the risk area.
Malaria risk varies by country and region, and this medicine is intended for travel to areas where malaria prophylaxis is recommended.

Ingredients
Main ingredients: Atovaquone 62.5 mg, proguanil hydrochloride 25 mg.
Excipients: Poloxamer 188, microcrystalline cellulose, low-substituted hydroxypropyl cellulose, povidone K30, sodium starch glycollate (Type A), magnesium stearate, hypromellose, titanium dioxide (E171), iron oxide red (E172), macrogol 400, polyethylene glycol 8000.
Usage and Instructions
This medicine is used for the prevention of malaria caused by Plasmodium falciparum in children.
The daily dose depends on body weight:
- 5 to 7.9 kg: half a tablet once daily
- 8 to 9.9 kg: Three-quarters of a tablet once daily
- 10 to 19.9 kg: one tablet once daily
- 20 to 29.9 kg: two tablets once daily
- 30 to 39.9 kg: three tablets once daily
- >40: One Malarone Adults tablet once daily
Treatment should start 24 to 48 hours before entering a malaria-endemic area, continue daily throughout the stay, and be continued for seven days after leaving the malaria risk area.
Tablets should be taken with food or a milky drink at the same time each day to improve absorption. If vomiting occurs within one hour of taking a dose, another dose should be taken as soon as possible.
This medicine reduces the risk of malaria but does not provide complete protection. Additional mosquito bite prevention measures should be used, and medical advice should be sought promptly if symptoms such as fever develop during or after travel.
Warnings
Do not give this medicine if the child is allergic to atovaquone, proguanil hydrochloride, or any of the other ingredients, or if the child has severe kidney disease.
This medicine is not recommended for malaria prevention in children weighing less than 5 kg.
Tell your pharmacist if the child is taking any other medicines, including treatments for nausea and vomiting, tuberculosis, HIV infection, blood clotting disorders, or infections, as some medicines may interfere with Malarone.
If the child becomes unwell, particularly with vomiting or diarrhoea, the effectiveness of malaria prevention may be reduced.
Stop giving the tablets and seek urgent medical advice if the child develops signs of a serious allergic reaction or a severe skin reaction.
Side Effects
Like all medicines, this medicine can cause side effects, although not everybody gets them.
Very common side effects include headache, nausea, vomiting, stomach pain, and diarrhoea.
Common side effects include dizziness, sleep disturbance, unusual dreams, loss of appetite, fever, cough, rash, and changes in blood test results affecting the liver or blood cells.
Uncommon side effects include anxiety, palpitations, mouth inflammation, hair loss, hives, and increased levels of amylase in blood tests.
Rare side effects include hallucinations.
Serious side effects requiring immediate medical attention include severe allergic reactions, serious skin reactions such as blistering or peeling, seizures, inflammation of the liver, and severe changes in blood cell counts.
